چکیده
Abstract Natural killer (NK) cells have been implicated in Ebola virus disease (EVD) pathogenesis as well protection against viruses. A clear understanding of their role during EVD is needed for better exploiting functionalities designing effective countermeasures Previously, we reported that protein VP40 induces IL-12 and IL-18 dependent activation human NK involves accessory cells. Here, extended our studies to define the identities molecular mechanism(s) underlying triggering cell activation. Purified CD14 +myeloid cells, CD3 +T or CD19 +B isolated from normal peripheral blood mononuclear (PBMCs), were co-cultured with purified CD56 +NK cells; cultures activated expression IFN-γ, granzyme B, perforin, surface CD107a was measured. small, but significant, increase observed when alone incubated VP40. Inclusion resulted significant additional status. Both soluble factors cell-cell contact mechanisms involved VP40-induced, In addition, a small subset bound directly triggered. Together, data decipher both independent induced activation, findings may help anti-Ebola countermeasures. Supported by funds Cincinnati Children's Research Foundation.

TLR7/8-mediated activation of human NK cells results in accessory cell-dependent IFN-gamma production.
NK cells express receptors that allow them to recognize pathogens and activate effector functions such as cytotoxicity and cytokine production. Among these receptors are the recently identified TLRs that recognize conserved pathogen structures and initiate innate immune responses. We demonstrate that human NK cells express TLR3, TLR7, and TLR8 and that these receptors are functional. TLR3 is ex...

Natural killer (NK) cells play a crucial role in regulating immune functions. Few studies have characterized canine NK cells. We previously demonstrated that canine peripheral blood lymphocytes (PBLs) with a low surface CD5 density (CD5lo) are considered a critical NK population. Natural killer cells in most mammals do not express T-cell markers, but canine CD5lo cells express surface molecules...
متن کاملRegulation of Ebola virus VP40 matrix protein by SUMO
The matrix protein of Ebola virus (EBOV) VP40 regulates viral budding, nucleocapsid recruitment, virus structure and stability, viral genome replication and transcription, and has an intrinsic ability to form virus-like particles. The elucidation of the regulation of VP40 functions is essential to identify mechanisms to inhibit viral replication and spread. Post-translational modifications of p...
متن کاملAssociation of ebola virus matrix protein VP40 with microtubules.
Viruses exploit a variety of cellular components to complete their life cycles, and it has become increasingly clear that use of host cell microtubules is a vital part of the infection process for many viruses. A variety of viral proteins have been identified that interact with microtubules, either directly or via a microtubule-associated motor protein. Here, we report that Ebola virus associat...
